- Explain the how different solvent ratios and gradient types effect the separation of components in a mixture and the peak shape of individual analytes.
- Explain the principles of liquid-liquid partitioning.
- Explain the effect of solvent polarity on retention times
- Explain the effects of isocratic and gradient methods on retention times and peak shape.
- Create a calibration curve using HPLC and validate the calibration using a validation standard (control) solution.
- Quantitate the amount of analyte in a mixture using a calibrated HPLC.
- Interpret chromatographic data to identify components of a mixture and determine their concentration.

- 4.2: High performance liquid chromatography
- High performance liquid chromatography (HPLC) makes use of a high pressure pump to deliver a mobile phase solvent at a uniform rates at pressures. The most obvious advantage of HPLC over gravity liquid chromatography is that samples can be separated much more quickly and with reproducible elution times.
