Fractionation typically begins with centrifugation of the lysate. Using low-speed centrifugation, one can remove cell debris, leaving a supernatant containing the contents of the cell. By using successively higher centrifugation speeds (and resulting g forces) it is possible to separate out different cellular components, like nuclei, mitochondria, etc., from the cytoplasm.

In order for the results of size exclusion separations to be meaningful, there can be no directed forces between the compounds being separated and the surface of the particles used as the stationary phase. Instead, the particles are prepared with well-characterized pore sizes.

Draw a separation chromatogram if a group of these three molecules go through this column. Hint: molecular size is directly related to the formula mass of the molecules. If the pore sizes change, what would happen to the calibration curve? What type of applications of size exclusion chromatography can you think of?

The pore is bigger than would actually occur and the molecules would never have sizes that big relative to the size of the bead. The size of the pore can be controlled to some degree during the synthesis of the bead. Based on your answers above, now draw a chromatogram showing the elution order of the three molecules and label the peaks.

It emphasizes the importance of a well-structured sampling plan, including choices about sample types (e.g., random, systematic), collection methods (e.g., grab, composite), and considerations of the population's nature (homogeneous or heterogeneous).
