Firefly Luciferases are extracted by differential centrifugation and purified by gel filtration.  Luciferins are substrates of luciferases . Firefly luciferin emits at 562 nm on reaction with oxygen, catalysed by luciferase in the presence of adenosine triphosphate (ATP) and magnesium ions.
